AstraZeneca and Ionis said eplontersen failed to statistically significantly reduce the risk of cardiovascular events and death compared with placebo in the CARDIO-TTRansform study of more than 1,400 patients with TTR cardiomyopathy. The event rate was 29% in the eplontersen group, not substantially different from 32% in the placebo group, while 81% of patients used a stabilizer, the standard treatment. Jefferies raised the possibility that the additional benefit of combining a silencer with a stabilizer may be smaller than previously assumed, while Alnylam analyzed that it could adjust the clinical design of its next-generation candidate nucresiran.
