Decline in DNA-regulating enzyme Dnmt3a triggers Caspase-8 activation New therapeutic target proposed for ischemic heart disease

A mechanism by which myocardial infarction reduces the DNA-regulating enzyme Dnmt3a, activating the Caspase-8 gene and triggering cardiomyocyte pyroptosis, has been identified for the first time. Dnmt3a acts as a switch that suppresses Caspase-8 gene expression; when this function is disrupted, the cell-death protein Gsdmd is activated in a cascade. The Dnmt3a/Caspase-8/Gsdmd regulatory axis identified in this study has been proposed as a new therapeutic target for ischemic heart disease.
